Procainamide Hydrochloride is a blocker of sodium channel and an inhibitor of DNA methyltransferase. It is an anti-arrhythmic agent. It is a medication used to manage and treat ventricular arrhythmias, supraventricular arrhythmias, atrial flutter/fibrillation, and Wolf-Parkinson-White syndrome. It is in the antiarrhythmic Agent Class 1A class of medications[3]. Oral procainamide was associated with a significant decrease in ICD (implantable cardioverter-defibrillator) therapies and ventricular arrhythmias, showing an acceptable profile of tolerability[4]. Treatment of endotoxemic animals with procainamide not only inhibited the increased levels of DNMT1 and 5-methylcytosine but also ameliorated neutrophil infiltration and superoxide production in the lung. Moreover, both DNMT (DNA methyltransferase) inhibitors procainamide and hydralazine improved hypotension, hypoglycemia, and multiple organ dysfunction of LPS-treated rats[5]. Procainamide-induced autoimmunity is characterized predominantly by an anti-histone and anti-denatured DNA immune response[6].
