Chiauranib 生物活性

描述 Chiauranib (CS2164) is a multi-target inhibitor taken orally that acts against tumor angiogenesis. It effectively targets angiogenesis-related kinases (VEGFR1, VEGFR2, VEGFR3, PDGFRα, and c-Kit), the mitosis-related kinase Aurora B, and the chronic inflammation-related kinase CSF-1R, with IC50 values between 1-9 nM, exhibiting potent anticancer effects[1].
体内研究

Chiauranib (CS2164; 0.5-40 mg/kg; oral administration; once daily; for 33 days or 43 days) treatment leads to significant regression or complete cessation of tumor growth at doses well tolerated when administered orally across various human tumor xenograft models, demonstrating wide-ranging and strong in vivo anti-tumor effects[1].

体外研究

Chiauranib (CS2164; 3 μM; 24 hours) induces G2/M cell cycle arrest and inhibits cell proliferation in tumor tissues by blocking Aurora B-mediated H3 phosphorylation[1].

Chiauranib (0.03-3 μM) exerts anti-angiogenic effects by inhibiting VEGFR/PDGFR phosphorylation, blocking ligand-dependent cell proliferation and capillary tube formation, and preventing vasculature development in tumor tissues[1].

Chiauranib (CS2164) suppresses CSF-1R phosphorylation, which in turn inhibits ligand-induced monocyte-to-macrophage differentiation and decreases the presence of CSF-1R+ cells in tumor tissues[1].
