IWP-4 生物活性

描述 IWP-4 is a potent Wnt pathway inhibitor with an IC50 of 25 nM, promoting the expression of cardiac indicators such as cardiac troponin I (CTNI) and bright cells expressing cardiac myosin heavy chain (MYHhi+). It leads to the emergence of beating foci (0.44±0.10 SEM beats per second), not observed in cultures without IWP-4 treatment. Flow cytometric analysis reveals a significant increase in MYHlo+ cells in IWP-4 treated cultures compared to untreated ones at day 16, with percentages of 17.0±1.3 SD% and 5.4±1.4 SD%, respectively. Additionally, 63% (481/817) of cells treated with IWP-4 show nuclear NKX2-5 protein expression[1]. Mesenchymal precursor cells (MPCs) exposed to IWP-4 do not exhibit significant differences in AXIN2, CTNNB1, and GSK3B expression levels compared to those treated with osteogenic medium alone on day 7. However, by day 21, MPCs treated with IWP-4 show increased levels of DKK1 and GSK3β. IWP-4 also significantly reduces the expression of SPARC and COL1A1[2].
