Oxytetracycline HCI 生物活性

描述 Oxytetracycline hydrochloride, a member of the tetracycline class of antibiotics, effectively inhibits both Gram-negative and Gram-positive bacteria. It functions as a protein synthesis inhibitor by preventing the binding of aminoacyl-tRNA to the mRNA-ribosome complex and also exhibits anti-HSV-1 (Herpes Simplex Virus-1) activity[1].[2].[3].

The pharmacological impact of oxytetracycline varies across species. When administered at a therapeutic dose of 82.8 mg/kg of body weight to 1% body weight per day for 10 days, it influences various biological parameters differently among fish species. In Morone chrysops x M. saxatilis, it increases relative liver weight. In Ictalurus punctatus, it enhances the enzymatic activity of CYP3A4, whereas in Oreochromis niloticus, it increases protein expression of CYP3A4 but depletes hepatic CYP3A4[1].

Regulatory limits for oxytetracycline residue include 100 μg/kg in muscle and milk, 200 μg/kg in eggs, 300 μg/kg in liver, and 600 μg/kg in kidney tissues. In aquaculture, oxytetracycline is typically administered via medicated feed at concentrations ranging from 35 to 75 mg active ingredient per kg of biomass per day for durations of 7 to 14 days[1].

Oxytetracycline is also a key member of the bacterial aromatic polyketide family, a class of structurally diverse natural products. It is synthesized via a type II polyketide synthase process, which constructs a poly-beta-ketone backbone through successive decarboxylative condensation of malonyl-CoA extender units. This backbone is then modified by various enzymes such as cyclases, oxygenases, transferases, and other tailoring enzymes to achieve its final structure[2].
