描述 | Wnt signaling is one of the key cascades regulating development and stemness, and has also been tightly associated with cancer[6]. IWP-4 is a small molecule Wnt inhibitor with an IC50 of 25 nM[7]. IWP-4 treatment resulted in a large percentage of cells expressing low amounts of cardiac myosin heavy chain and expression of early cardiac progenitor markers ISL1 and NKX2-5, thus indicating the production of large numbers of immature cardiomyocytes (~65,000/cm2 or ~1.5 per input human embryonic stem cell (hESC)). In addition, IWP-4 induced expression of both atrial (MLC2a) and ventricular (MLC2v) forms[7]. Mesenchymal precursor cells (MPCs) treated with IWP-4 express elevates levels of DKK1 (Dickkopf homolog 1) and GSK3β (glycogen synthase kinase 3 β) on day 21. IWP-4 also causes a significant down regulation of SPARC (secreted protein acidic and rich incysteine) and COL1A1 (Collagen 1A1)[8]. |
