NFAT Transcription Factor Regulator-1 inhibited IL-2 synthesis with an IC50 value of 182 nM. NFAT Transcription Factor Regulator-1 inhibited human and rat PBMC proliferation with IC50 values of 82 nM and 146 nM, respectively. NFAT Transcription Factor Regulator-1 inhibits the production of IL-4 and IL-5 in human T-cell lines, and has a similar effect on IL-2 release[1].

Animal study
The inhibitory potency of NFAT Transcription Factor Regulator-1 is approximately 10 times that of cyclosporine, and the inhibitory effects on T cell IL-2 production were comparable to those of NFAT Transcription Factor Regulator-1 and cyclosporine at doses of 3.0 mg/kg and 30 mg/kg, respectively. NFAT Transcription Factor Regulator-1 and cyclosporine at doses of 3.0 mg/kg and 30 mg/kg, respectively, had comparable inhibitory effects on T cell IL-2 production. The effect of blocking T cell cytokine production in monkeys suggests that NFAT Transcription Factor Regulator-1 has similar potential for transplantation as cyclosporine[1].
