AZD1283 is a potent antagonist of the P2Y12 receptor, possessing a binding IC50 of 11 nM and a GTPγS IC50 of 25 nM, illustrating its significant capability to prevent platelet aggregation. This characteristic renders AZD1283 a candidate for thromboembolic disorder research^[1].^[2].

体内研究

Despite its poor stability in rat liver microsomes (T1/2 = 6.08 min), AZD1283 shows better stability in dog (T1/2 = 201 min) and human liver microsomes (T1/2 = 65.0 min)^[1].

体外研究

AZD1283 exhibits excellent antiplatelet aggregation potency with an IC50 value of 3.6 μM^[1].

AZD1283 also shows substantial inhibitory action against various CYP450 enzymes, with IC50 values of 6.62 μM for CYP2C9, 0.399 μM for CYP2C19, and 4.28 μM and 3.64 μM for CYP3A4 (using Midazolam and Testosterone as substrates, respectively)^[1].

AZD1283 is noted to increase blood flow and reduce ADP-induced platelet aggregation, achieving an antithrombotic EC50 of 3 μg/(kg×min)^[2].
