描述 | Benproperine is an ARPC2 (Actin-related protein 2/3 complex subunit 2) inhibitor which works as an inhibitor of cancer cell migration and an anti-metastatic agent. Benproperine could bound directly to ARPC2 in cells and inhibit Arp2/3 function by displaying disruption of lamellipodial structure and inhibition of actin polymerization. Unlike Arp2/3 inhibitors, Benproperine selectively inhibited the migration of cancer cells but not normal cells. Benproperine significantly inhibited the migration, but not viability, of various types of cancer cells with IC50 values of 1-2 mM for DLD-1 and AsPC-1 cells. Oral administration of 50mg/kg Benproperine 5 days per week for 4 weeks strongly suppressed lung metastasis of AsPC-1 cells (56.1% inhibition) and metastasis of HCT-116 cells by 78.9% and DLD-1 cells by 78.2%. | ||
