描述 Protein photocrosslinking, via the introduction of non-natural amino acids, has emerged as a superior strategy for the covalent trapping of transient protein-protein interactions in vivo, which is generally not feasible using other methodologies. This method is particularly suitable for detecting periplasmic proteins interacting with HdeA at a low pH. DiZPK, an alkyl diazirine–containing unnatural amino acid, is a new photocrosslinker for identifying direct protein-protein interactions in living prokaryotic and eukaryotic cells[2].
作用机制 Introduction of DiZPK at two dimer-interface sites, Phe35 and Ala38, yielded a crosslinking efficiency[2].
