描述 CHAPS is a non denaturing zwitterionic detergent for membrane biochemistry. It can be used to dissolve membrane proteins and interrupt protein-protein interactions. The zwitterionic detergent CHAPS, a derivative of the bile salts, is widely used in membrane protein solubilization. CHAPS has a low affinity for the erythrocyte membrane (partition coefficient K=0.06mM(-1)), and at sub-hemolytic concentrations it causes little effect on membrane lipid order. CHAPS hemolysis and phospholipid solubilization are closely correlated[1]. Chemical cross-linking shows that the dimerization of EnvZ-PD is significantly inhibited by the CHAPS and cholate[2]. The recovery and preservation of catalytic activity of two trans-membrane proteases, dipeptidyl peptidase IV and nephrilysin, was examined and found to be clearly superior after CHAPS treatment compared to DTT(dithiothreitol). Proteomic profiling by mass spectrometry (MS) revealed that 76.2% of proteins recovered by CHAPS are common to those seen for DTT treatment, which illustrates underlining similarities between the two approaches[3].
