描述 | Wnt/β-catenin signaling pathway plays crucial roles in multiple stages of development and tissue homeostasis, and its dysregulation can lead to various human diseases. MSAB is a selective inhibitor of Wnt/β-catenin signaling. In HCT116 cells, MSAB resulted in strong inhibition of the TOP-Luc activity in a dose dependent manner. MSAB inhibited proliferation of Wnt-dependent cancer cell lines at concentrations as high as 10 µM. In vivo, MSAB (10 or 20 mg/kg; i.p.; daily for 2 weeks) markedly reduced the size and weight of various types of Wnt-dependent HCT116, HT115, and H23 tumors in xenografted mouse models. Moreover, there was an increase in the level of cleaved caspase 3 in HCT116 and H23 xenograft tumor tissues from mice treated with MSAB, demonstrating that MSAB treatment resulted in apoptotic cell death of tumor xenograft cells leading to subsequent reduction in tumor size[1]. | ||
