描述 | Eudesmin, a natural product isolated and purified from the dry leaves of Haplophyllum perforatum Kar.et Kir. with antiinflammatory effects, can significantly inhibit TNF-alpha production by lipopolysaccharide (LPS)-stimulated murine macrophage RAW264.7 without displaying cytotoxicity, and can significantly attenuate T cell proliferation stimulated by concanavalin A (Con A) in a dose-dependent manner. Eudesmin can induce vascular relaxation in rat aorta, which is mediated by release of nitric oxide and prostanoid through the involvement of histamine receptor present in the endothelial cells, and can induce neurite outgrowth and enhance nerve growth factor (NGF)-mediated neurite outgrowth from PC12 cells by stimulating up-stream MAPK, PKC and PKA pathways at a concentration of 50 mM, showing neuritogenic activity. Eudesmin also has significant anticonvulsant and sedative effects, and the mechanism of eudesmin may be related to up-regulation of GABAA and GAD65 expressions, and anti-apoptosis of neuron the in brain. |
