描述 The root of Paeonia lactiflora Pall (family Ranunculaceae) or peony root is a commonly used Chinese herbal medicine in clinical practice. Empirical evidence suggests that various peony root preparations have broad therapeutic benefits for pain, muscle spasm, inflammation, menstrual dysfunction, and neurodegenerative disorder. The isomer albiflorin (AF) is a major constituent contained in peony root. Pretreatment with AF at 50 μM and 100 μM significantly reduced the lethal effect of Glu (P<0.05). Pretreatment with 100 μM AF almost completely inhibited Glu-induced apoptotic alternation, with a >93 % of survival rate compared to 84 % in untreated cells (P<0.05). Moreover, Glu-induced increase of ROS level was suppressed to control level by 100 μM AF pretreatment (P<0.05), suggesting that the protective effect of AF against Glu neurotoxicity are associated with the reduction of intracellular ROS level. In the presence of 100 μM AF, the phosphorylation of AKT and GSK3β was up-regulated in a time-dependent manner (P<0.05) indicating that the activation of AKT/GSK-3β is associated with the protective effects of AF against Glu neurotoxicity[3].
