描述 | Oxythiamine (OT), an analogue of anti-metabolite, is an inhibitor of transketolase, and a thiamine antagonist. Oxythiamine can suppress the nonoxidative synthesis of ribose and induce cell apoptosis by causing a G1 phase arrest in vitro and in vivo. It altered dynamics of protein signals by suppressing phosphor kinase expression in a concentration (0μM, 5μM, 50μM and 500μM)-dependent and time (0h, 12h and 48h)-dependent manner in MIA PaCa-2 cells. A time-dependent, but not dose-dependent, down-regulation of Annexin A1 expression was observed. The inhibition of transketolase by Oxythiamine resulted in changes of multiple cellular signaling pathways associated with cell apoptosis. |
