描述 | DTT is a reducing agent. DTT forms a stable six-membered ring with an internal disulfide bond once oxidized. DTT is widely used in biochemistry works to reduce dissulfide bridges, protect biomolecules, in sample preparation, and to denature proteins before electrophoresis analysis (SDS-PAGE). The thiol-containing compound Dithiothreitol (DTT) has been shown to be toxic to cultured cells by inducing the generation of reactive oxygen species that ultimately cause cell death[1]. Daratumumab (DARA) consistently interferes with routine blood bank serologic testing by directly binding to CD38 expressed on reagent red blood cells (RBCs). Treating RBCs with dithiothreitol (DTT) eliminates the DARA interference. The DTT method is robust and reproducible and can be implemented by transfusion services worldwide to help provide safe blood products to patients treated with DARA (Daratumumab)[2]. The dithiothreitol (DTT) assay is the most commonly used method to quantify the oxidative potential of fine particles[3]. |
