描述 Pitstop 2 is a clathrin inhibitor that inhibits clathrin-mediated endocytosis (CME) by binding to the terminal structural domain of clathrin[1][2].At a concentration of 20-40 μM for 30 minutes, pitstop 2 can inhibit the endocytosis of transferrin by J774A.1 macrophages without damaging cell viability or affecting cholera toxin B internalization in J774A.1 macrophages[1].At concentrations less than 100 μM for 6 hours, pitstop 2 inhibits the mitotic spindle and affects mitotic progression in HeLa cells. In the concentration range of 1-30 μM, pitstop 2 induced apoptosis in dividing cancer cells and inhibited cell growth at 24 h. At 48 h, pitstop 2 inhibited the mitotic spindle and affected the mitotic progression of HeLa cells. At 48 hours of action, pitstop 2 did not affect the growth and viability of non-carcinogenic NIH3T3 fibroblasts[2].
