描述 | Anandamide (arachidonoylethanolamide, AEA) is endogenous ligands of cannabinoid CB1 and CB2 receptors and AEA is metabolized by the enzyme fatty acid amide hydrolase (FAAH). Biochanin A (4-Methylgenistein; Olmelin), a major isoflavone in red clover, is a naturally occurring FAAH inhibitor, with IC50 values of 1.8, 1.4 and 2.4 µM for mouse, rat and human FAAH ,respectively[3]. In vitro, Biochanin A inhibited human osteosarcoma cell growth and colony formation in a dose-dependent manner, and induced apoptosis by triggering activation of the intrinsic mitochondrial pathway and caspase-9 and -3[4]. In vivo, type 2 diabetes mellitus animals treated with 10, 20 and 40 mg/kg of Biochanin A for 28 days showed significantly decreased blood glucose, reduced insulin resistance and improved inulin sensitivity, compared to model group. These effects might be linked to its effects on SIRT1[5]. |
