描述 3-Methyltoxoflavin is a potent PDI inhibitor with an IC50 value of 170 nM. 3-Methyltoxoflavin is toxic to human glioblastoma cell lines. Bru-seq analysis showed that 3-Methyltoxoflavin induced Nrf2 antioxidant response, ER stress response and autophagy. Specifically, 3-Methyltoxoflavin upregulated the transcription and protein expression of heme oxygenase 1 and SLC7A11, and inhibited PDI target genes such as TXNIP and EGR1. Interestingly, 3-Methyltoxoflavin-induced cell death is not through apoptosis or necrosis, but through a hybrid process of autophagy and iron metamorphosis[1].
